Hello. It is first time to work with NMR assignment and sparky.

I searched previous conversations but there was no appropriate subjects for me.


Recently I am working with 22 a.a peptide.

I finished backbone assignment manually.

N-HSQC, HNCACB, HNCOCACB, HNCO, HNCACO -these 5 spectra were used.


But.. the problem is side-chain assignment.

I only have C-HSQC, HC(c)H-TOCSY, (h)CCH-TOCSY, hCCH-COSY, N,C-NOESY

I dont know how to approach with this spectra.



1. Axis is confusing. w1 w2 w3. I input xa-xa-xx as I did with HNCACB. etc. Is that right?

I will attach image file.

2. Do you recommend automatic or manual?

3. Is another spectra needed for side chain assignment? HCCH is enough for side chain assignment ? please consider this peptide is short: 22 a.a
