Hi Lee

I have got about 90% assignment from my 3D triple resonance backbone experiments for my mutant protein after run on PINE-Server3.0 . But the assignment looks quite different from my Wild protein. So, I would like to refine the assignment for my mutant protein with my pre -assignment from my Wild protein assignment. And then I would like to compare results between the both assignment ( refined with wild protein assignment) (without wild protein assignment).

Regards

You still need to provide triple resonance experiments to run PINE even though you are providing pre-assignment file. If you just simply compare, you can use ta to deploy peaks onto N-HSQC. Given your information, it is difficult to know what exact thing you would like to do. Is your reference assignment is from the same sequence protein? Do you want to compare HSQCs from different conditional states?
